Wide-Field Microscopy
The pixel size here is fixed after the objective lens is selected.
Laser Scanning Confocal Microscopy
The pixel size here varies on the scanning resolution.

In a 2D image, the area can be calculated as the product of the pixel number and the area of one pixel.
