Wang et al. lists the following steps for performing cryo electron tomography:

  1. Cells are grown on the EM grids.
  2. The cells are plunged-freezed in liquid ethane or propane.
  3. Cryo FIB milling is done on an Aquilos to make the sample thin enough for TEM.
  4. Cryo tomography data is done on the Krios TEM.
  5. Data analysis, segmentation, and sub-tomogram averaging is done.
