10.3 Cryo Electron Tomography in situ
Wang et al. lists the following steps for performing cryo electron tomography:
- Cells are grown on the EM grids.
- The cells are plunged-freezed in liquid ethane or propane.
- Cryo FIB milling is done on an Aquilos to make the sample thin enough for TEM.
- Cryo tomography data is done on the Krios TEM.
- Data analysis, segmentation, and sub-tomogram averaging is done.
